How Pregabalin (LYRICA) Works?


Mechanism of action:

 Mechanism of action of pregabalin has not been exactly explained. It binds with high affinity to the alpha2-delta site - subunit of voltage-dependent calcium channels in central nervous system.Thus, calcium currents decrease, which may be associated with the antinociceptive and antiseizure effects of pregabalin. Nociception is the recognizing of painful stimuli.



What pregabalin is indicated for?

Pregabalin (Brand name: Lyrica)  is indicated for neuropathic pain, concomitant with diabetic peripheral neuropathy,  postherpetic neuralgia, fibromyalgia and  as adjunctive therapy for adult patients with partial onset seizures. 


What are the contraindications for the use of pregabalin?

 It contraindicated in cases with known hypersensitivity to pregabalin. Angioedema and hypersensitivity  reactions included skin redness, blisters, hives, rash, dyspnea, and wheezing have occurred. 


What are the side effects of taking pregabalin?

Peripheral Edema. Dizziness & Somnolence.
Weight Gain.
Tumorigenic Potential.
Ophthalmological Effects: blurred vision.
Creatine Kinase Elevations.
Decreased Platelet Count.
PR Interval Prolongation.
Abuse. Dependence. Insomnia, nausea, headache or diarrhea following abrupt or rapid discontinuation of Lyrica.


Use of pregabalin in pregnant women, nursing mothers and children. 

Increased incidences of fetal structural abnormalities were observed in the offspring of rats and rabbits given pregabalin during pregnancy, at high doses.  There are no adequate studies in pregnant women.Thus use LYRICA during pregnancy only if the potential benefit justifies the potential risk to the fetus. This also applies to nursing mothers children.
